As a rule most plugins can be deactivated, removed and re-installed as settings are not stored in the plugin folder, they are stored in the database or somewhere else.
Also the latest version of WordPress allows plugins to be installed on top of an older version, you get a confirmatioin before it goes ahead.
But of course it never hurts to backup before making any changes 🙂
I sometimes compress a plugin folder to a zip file in my cPanel just to be on the safe side!
